Background
FOXF1 (Forkhead Box F1) is a transcription factor belonging to the forkhead box (FOX) family of proteins, characterized by a conserved DNA-binding domain known as the forkhead domain. This protein plays a critical role in embryonic development, particularly in the formation of the lungs, gastrointestinal tract, and cardiovascular system, where it regulates cell proliferation, differentiation, and tissue-specific gene expression. FOXF1 functions as both an activator and repressor of transcription, interacting with various co-factors and signaling pathways, including TGF-β and Wnt signaling, to modulate target gene expression. Beyond development, FOXF1 has emerged as a significant player in cancer biology, where its expression is frequently dysregulated; it can act as either an oncogene or tumor suppressor depending on the tissue context, influencing processes such as epithelial-mesenchymal transition (EMT), angiogenesis, and metastasis. Mutations or deletions in the FOXF1 gene are associated with Alveolar Capillary Dysplasia with Misalignment of Pulmonary Veins (ACD/MPV), a rare and often fatal congenital lung disorder, highlighting its essential role in pulmonary vascular development. Additionally, FOXF1 is implicated in various other pathological conditions, including liver fibrosis, gastrointestinal disorders, and certain malignancies such as colorectal cancer and rhabdomyosarcoma, making it an important subject of ongoing biomedical research for potential therapeutic targeting.
